Position Summary
Matchpoint seeks a highly motivated scientist with a strong background in mass spectrometry-based proteomics and protein biochemistry to be an integral member of our chemoproteomics and chemical biology team. In this role, you will be responsible for operating Thermo Orbitrap and Agilent TOF Mass Spectrometer(s) and will lead the design and execution of intact mass, peptide mapping, and binding kinetics studies to characterize covalent molecules. The desired candidate will collaborate with internal and external scientists in addressing key scientific questions and be an integral member of a multidisciplinary team. We are seeking a qualified candidate who will share our excitement about the science and contribute to our culture.

Key Responsibilities

  • Lead the design, optimization and execution of recombinant protein intact mass and peptide mapping as screening and confirmatory experiments to discover novel drug targets and develop covalent therapeutics to treat immunological disorders.
  • Maintain and oversee complex laboratory equipment including Thermo Fisher Orbitrap mass spectrometers (Eclipse and Exploris), nano-flow high performance liquid chromatography (nHPLC) systems, Agilent TOF
  • Develop data dependent acquisition (DDA) MS methods and targeted proteomics-based methods.
  • Conduct protein/peptides target-ligand interactions identification (intact mass and peptide mapping), and mechanistic/binding kinetics evaluation using proprietary and commercially available bioinformatics software (using the SEQUEST algorithm and other bioinformatics packages such as MaxQuant, Skyline, MassPike, Proteome Discoverer, Agilent BioConfirm, Thermo Biopharma Finder and/or ProSightPD).
  • Maintain high-quality electronic laboratory notebook and reporting of data.
  • Write and standardize protocols used for all mass spectrometry-based workflows and methods maintain records to industry standards

Key Qualifications

  • Ph.D. in Chemistry, Chemical Biology, Biochemistry, Enzymology, or related field with a focus on mass spectrometry.
  • 3-5 years of industry experience in nano-scale proteomics methodology employing Thermo Fisher Orbitrap / Agilent TOF mass spectrometers.
  • Prior expertise in characterization of small molecules by peptide mapping, post-translational modification assignment, disulfide mapping, intact mass analysis and binding kinetics determination.
  • Knowledge of sample preparation, and the fundamentals of chromatography and mass spectrometry are needed as the candidate is expected to identify assay issues and perform the associated troubleshooting.
  • Hands on experience in operation, maintenance & trouble shooting of Thermo Fisher Orbitrap and Agilent TOF LC-MS instruments and associated software (MaxQuant, Skyline, MassPike, Proteome Discoverer, Agilent BioConfirm, Thermo Biopharma Finder and/or ProSightPD)
  • Ability to work collaboratively with multiple groups on technical projects exhibiting self-initiative and drive for results.
  • Strong technical competency and well organized approach to research
  • Excellent oral and written communication skills.
  • Hands-on development of LC/MS/MS methods to quantify peptide and protein therapeutics
